A significant number of wells show still clear drops after precipitant additon. In situ DLS analysis is quite enlightening. There are recurring distribution patterns which can be subdivided into classes. Among these classes is a certain signature that indicates the presence of liquid dense cluster, a step in the homogenious nucleation mechanism of many proteins.
Hanging Drop Nucleation detection
In situ DLS can be used to detect particle sizes direktly in hanging drop crystallization trials. Depending on the added preciciptant concentration the response of the protein is different according to the position in the phase diagram.
Mother Liquer Size Distribution Analysis
In situ DLS analysis in drops containing growing crystals. Size distribution plots reveal a mixture of particles with a huge size variety. Nevertheless the monomeric state that enables crystal growth could be detected in all drops.
